I've had similar experiences.
It depends what the problem is.
Sometimes you can ignore the assistant coach and play anyways (which I've done many times by just pressing continue on the preview play game screen).
At times the problem is that there are not enough defence or offense players to create a lineup. You might have 23 players but not enough defence players or not enough offense players.
