Pinch Hitting from Depth Chart

When you're setting your lineup/depth chart, you have a spot to insert pinch hitters. My question is:

If I'm benching some lefty bats when facing a LHP, should I insert these guys as pinch hitters in the depth chart for vs RHP or LHP? I'm assuming vs RHP but wasn't sure if the game only looked at that at the start of a game.

I'm hoping my thinking is correct since the game definitely checks for defensive subs (which obviously happen later in a game).
